The interacting effects of global changes—including increased temperature, altered precipitation, reduced acidification and increased dissolved organic matter loads to lakes—are anticipated to create favourable environmental conditions for cyanobacteria in northern lakes. However, responses of cyanobacteria to these global changes are complex, if not contradictory. We hypothesized that absolute and relative biovolumes of cyanobacteria (both total and specific genera) are increasing in Swedish nutrient‐poor lakes and that these increases are associated with global changes. We tested these hypotheses using data from 28 nutrient‐poor Swedish lakes over 16 years (1998–2013). Increases in cyanobacteria relative biovolume were identified in 21% of the study sites, primarily in the southeastern region of Sweden, and were composed mostly of increases from three specific genera: Merismopedia, Chroococcus and Dolichospermum. Taxon‐specific changes were related to different environmental stressors; that is, increased surface water temperature favoured higher Merismopedia relative biovolume in low pH lakes with high nitrogen to phosphorus ratios, whereas acidification recovery was statistically related to increased relative biovolumes of Chroococcus and Dolichospermum. In addition, enhanced dissolved organic matter loads were identified as potential determinants of Chroococcus suppression and Dolichospermum promotion. Our findings highlight that specific genera of cyanobacteria benefit from different environmental changes. Our ability to predict the risk of cyanobacteria prevalence requires consideration of the environmental condition of a lake and the sensitivities of the cyanobacteria genera within the lake. Regional patterns may emerge due to spatial autocorrelations within and among lake history, rates and direction of environmental change and the niche space occupied by specific cyanobacteria.  相似文献

Fayetteville Green Lake (FGL) is a recognized, extensively studied present‐day model of the stratified Proterozoic ocean. Nonetheless, biomass sedimentation in FGL remains hard to explain: while virtually all sediment pigments belong to photosynthetic sulfur bacteria from a chemocline, the isotopic carbon signature of the bulk organic matter suggests its epilimnetic phytoplankton origin. To explain the epilimnetic origin of sedimented carbon, we studied the dominant Synechococci, isolated from FGL. Here, we present experimental evidence that FGL Synechococci produce copious extracellular polysaccharides (EPS) especially when availability of inorganic carbon (Ci) is high relative to availability of other macronutrients, for example phosphorus. The accumulating EPS become impregnated with calcium, magnesium, and sodium cations and are released to the environment as ballasted cell coverings. Sedimentation of these cell‐free EPS can constitute the bulk of pigment‐free organic material in FGL sediment. Because increased availability of Ci specifically stimulates production of EPS and the accumulated EPS adsorb cations and become ballasted, we propose the universal role of cyanobacterial EPS in biomass sedimentation in the high‐Ci Paleoproterozoic ocean as well as in modern aquatic systems like FGL.  相似文献

Photosynthetic generation of reducing power makes cyanobacteria an attractive host for biochemical reduction compared to cell‐free and heterotrophic systems, which require burning of additional resources for the supply of reducing equivalent. Here, using xylitol synthesis as an example, efficient uptake and reduction of xylose photoautotrophically in Synechococcus elongatus PCC7942 are demonstrated upon introduction of an effective xylose transporter from Escherichia coli (Ec‐XylE) and the NADPH‐dependent xylose reductase from Candida boidinii (Cb‐XR). Simultaneous activation of xylose uptake and matching of cofactor specificity enabled an average xylitol yield of 0.9 g g?1 xylose and a maximum productivity of about 0.15 g L?1 day?1 OD?1 with increased level of xylose supply. While long‐term cellular maintenance still appears challenging, high‐density conversion of xylose to xylitol using concentrated resting cell further pushes the titer of xylitol formation to 33 g L?1 in six days with 85% of maximum theoretical yield. While the results show that the unknown dissipation of xylose can be minimized when coupled to a strong reaction outlet, it remains to be the major hurdle hampering the yield despite the reported inability of cyanobacteria to metabolize xylose.  相似文献

In oxygenic photosynthetic organisms, excluding angiosperms, flavodiiron proteins (FDPs) catalyze light‐dependent reduction of O2 to H2O. This alleviates electron pressure on the photosynthetic apparatus and protects it from photodamage. In Synechocystis sp. PCC 6803, four FDP isoforms function as hetero‐oligomers of Flv1 and Flv3 and/or Flv2 and Flv4. An alternative electron transport pathway mediated by the NAD(P)H dehydrogenase‐like complex (NDH‐1) also contributes to redox hemostasis and the photoprotection of photosynthesis. Four NDH‐1 types have been characterized in cyanobacteria: NDH‐11 and NDH‐12, which function in respiration; and NDH‐13 and NDH‐14, which function in CO2 uptake. All four types are involved in cyclic electron transport. Along with single FDP mutants (?flv1 and Δflv3) and the double NDH‐1 mutants (?d1d2, which is deficient in NDH‐11,2 and ?d3d4, which is deficient in NDH‐13,4), we studied triple mutants lacking one of Flv1 or Flv3, and NDH‐11,2 or NDH‐13,4. We show that the presence of either Flv1/3 or NDH‐11,2, but not NDH‐13,4, is indispensable for survival during changes in growth conditions from high CO2/moderate light to low CO2/high light. Our results show functional redundancy between FDPs and NDH‐11,2 under the studied conditions. We suggest that ferredoxin probably functions as a primary electron donor to both Flv1/3 and NDH‐11,2, allowing their functions to be dynamically coordinated for efficient oxidation of photosystem I and for photoprotection under variable CO2 and light availability.  相似文献

Restoration targets for biological soil crusts are largely unknown. We surveyed seven 80‐year‐old grazing exclosures across northern Nevada for biocrusts to quantify reference conditions at relatively undisturbed sites. Exclosures were associated with the following plant communities: Wyoming big sagebrush, black sagebrush, and areas co‐dominated by winterfat and Wyoming big sagebrush. Cover of biocrusts and shrubs were generally higher than other plant groups at these sites, regardless of being inside or outside of the exclosures, suggesting these groups make up most of the native flora across the region. Important in forming soil structure, cyanobacteria of the order Oscillatoriales were less abundant and diverse in black sagebrush communities. Grazing had a negative effect on the abundance of Oscillatoriales but not the number of algal taxa, including cyanobacteria. Abundance of light algal crusts were not influenced by plant community or grazing. Dark algal crusts were generally less abundant on grazed sites. Influences of plant community and grazing were most apparent when accounting for reproductive rates of lichens and mosses based on establishment mechanisms. Abundance of shrubs, perennial grasses, Oscillatoriales, fast reproducing biocrusts and the number of algal and cyanobacterial taxa, varied by plant community, suggesting that restoration should be plant community specific. We demonstrate the affinity of rapidly reproducing biocrusts for winterfat‐Wyoming big sagebrush co‐dominated plant communities, regardless of grazing pressure. Across sites, the effects of grazing were most evident on the abundance of Oscillatoriales and slowly reproducing biocrusts following 80 years of cessation from grazing.  相似文献

Synechocystis sp. PCC 6803, a cyanobacterium widely used for basic research, is often cultivated in a synthetic medium, BG-11, in the presence of 4-(2-hydroxyethyl)-1-piperazine ethanesulfonic acid (HEPES) or 2-[[1,3-dihydroxy-2-(hydroxymethyl)propan-2-yl]amino]ethanesulfonic acid buffer. Owing to the high cost of HEPES buffer (96.9% of the total cost of BG-11 medium), the biotechnological application of BG-11 is limited. In this study, we cultured Synechocystis sp. PCC 6803 cells in BG-11 medium without HEPES buffer and examined the effects on the primary metabolism. Synechocystis sp. PCC 6803 cells could grow in BG-11 medium without HEPES buffer after adjusting for nitrogen sources and light intensity; the production rate reached 0.54 g cell dry weight·L−1·day−1, exceeding that of commercial cyanobacteria and Synechocystis sp. PCC 6803 cells cultivated under other conditions. The exclusion of HEPES buffer markedly altered the metabolites in the central carbon metabolism; particularly, the levels of compatible solutes, such as sucrose, glucosylglycerol, and glutamate were increased. Although the accumulation of sucrose and glucosylglycerol under high salt conditions is antagonistic to each other, these metabolites accumulated simultaneously in cells grown in the cost-effective medium. Because these metabolites are used in industrial feedstocks, our results reveal the importance of medium composition for the production of metabolites using cyanobacteria.  相似文献

The ability to cultivate the cyanobacterium Arhtrospira platensis in artificially lightened photobioreactors using high energetic efficiency (quasi‐monochromatic) red LED was investigated. To reach the same maximal productivities as with the polychromatic lightening control conditions (red + blue, P/2e? = 1.275), the need to work with an optimal range of wavelength around 620 nm was first established on batch and continuous cultures. The long‐term physiological and kinetic behavior was then verified in a continuous photobioreactor illuminated only with red (620 nm) LED, showing that the maximum productivities can be maintained over 30 residence times with only minor changes in the pigment content of the cells corresponding to a well‐known adaptation mechanism of the photosystems, but without any effect on growth and stoichiometry. For both poly and monochromatic incident light inputs, a predictive spectral knowledge model was proposed and validated for the first time, allowing the calculation of the kinetics and stoichiometry observed in any photobioreactor cultivating A. platensis, or other cyanobacteria if the parameters were updated. It is shown that the photon flux (with a specified wavelength) must be used instead of light energy flux as a relevant control variable for the growth. The experimental and theoretical results obtained in this study demonstrate that it is possible to save the energy consumed by the lightening device of photobioreactors using red LED, the spectral range of which is defined according to the action spectrum of photosynthesis. This appears to be crucial information for applications in which the energy must be rationalized, as it is the case for life support systems in closed environments like a permanent spatial base or a submarine. © 2009 American Institute of Chemical Engineers Biotechnol. Rising atmospheric CO2 concentrations are predicted to have a significant impact on global phytoplankton populations. Of particular interest in freshwater systems are those species that produce toxins or impact water quality, though evidence for how these species, and many others, will respond is limited. This study investigated the effects of elevated CO2 (1,000 ppm) relative to current atmospheric CO2 partial pressures (400 ppm), on growth, cell size, carbon acquisition, and photophysiology of five freshwater phytoplankton species including a toxic cyanophyte, Raphidiopsis raciborskii, from Lake Wivenhoe, Australia. Effects of elevated CO2 on growth rate varied between species; notably growth rate was considerably higher for Staurastrum sp. and significantly lower for Stichococcus sp. with a trend to lower growth rate for R. raciborskii. Surface area to volume ratio was significantly lower with elevated CO2, for all species except Cyclotella sp. Timing of maximum cell concentrations of those genera studied in monoculture occurred in the lake in order of CO2 affinity when free CO2 concentrations dropped below air equilibrium. The results presented here suggest that as atmospheric levels of CO2 rise, R. raciborskii may become less of a problem to water quality, while some species of chlorophytes may become more dominant. This has implications for stakeholders of many freshwater systems.  相似文献

The role of dissolved inorganic phosphorus (DIP) in promoting dominance of the toxic nitrogen (N)‐fixing cyanobacterium Cylindrospermopsis raciborskii (Wo?osz.) Seenayya et Subba Raju was examined in a subtropical water reservoir, Lake Samsonvale (=North Pine reservoir). A novel in situ bioassay approach, using dialysis tubing rather than bottles or bags, was used to determine the change in C. raciborskii dominance with daily additions of DIP. A statistically significant increase in dominance of C. raciborskii was observed when DIP was added at two concentrations (0.32 μM and 16 μM) in a daily pulse over a 4 d period in three separate experiments in the summer of 2006/2007. There was an increase in both C. raciborskii cell concentrations and biovolume in two DIP treatments, but not in the ammoniacal N + DIP treatment. In addition, overall phytoplankton cell concentrations increased with DIP addition, indicating that Lake Samsonvale was DIP limited at the time of experiments. Given the bioassay response, it is likely that dominance of C. raciborskii could increase in Lake Samsonvale with periodic injections of DIP such as inflow events.  相似文献
